چکیده
Artificial enzymes utilizing the genetically encoded non-proteinogenic amino acid p-aminophenylalanine (pAF) as a catalytic residue are able to react with carbonyl compounds through an iminium ion mechanism promote reactions that have no equivalent in nature. Herein, we report vivo biocatalytic cascade is augmented such artificial enzyme-catalysed new-to-nature reaction. The enzyme this study pAF-containing evolved variant of lactococcal multidrug-resistance regulator, designated LmrR_V15pAF_RMH, which efficiently converts benzaldehyde derivatives produced into corresponding hydrazone products inside E. coli cells. These cascades comprising artificial-enzyme-catalysed reaction important step towards achieving hybrid metabolism.

متن کاملBaker’s yeast catalyzed Henry reaction: Biocatalytic C-C bond formation
The C-C bond formation is an important reaction in organic synthesis to obtain value-added intermediates. Therefore, in this paper an attempt has been made to accelerate the Henry reaction (C-C bond formation) between aryl aldehydes and nitromethane using less expensive whole cell biocatalyst, baker’s yeast (BY). The scope of the methodology was also tested for the heteryl aldehyde i.e. 2-chlor...
متن کاملBiocatalytic Friedel–Crafts Acylation and Fries Reaction
The Friedel-Crafts acylation is commonly used for the synthesis of aryl ketones, and a biocatalytic version, which may benefit from the chemo- and regioselectivity of enzymes, has not yet been introduced. Described here is a bacterial acyltransferase which can catalyze Friedel-Crafts C-acylation of phenolic substrates in buffer without the need of CoA-activated reagents. Conversions reach up to...

متن کاملControlling the direction of photocurrents by means of CdS nanoparticles and cytochrome c-mediated biocatalytic cascades.
Cathodic or anodic photocurrents are generated by a monolayer of CdS nanoparticles in the presence of the oxidized or reduced states of cytochrome c, respectively, and the photocurrents are amplified by enzyme-generated biocatalytic cascades mediated by cytochrome c.
